Compression and drug release characteristics of KLUCEL HPC controlled release matrix systems

by Hercules Inc.

This white paper by Hercules Inc. explains compression and drug release characteristics of directly compressible KLUCEL hydroxypropylcellulose (HPC) controlled release matrix systems as a directly compressible matrix polymer in combination with various dilutents. HPC can be used as a controlled release tablet matrix, and reducing the particle size of HPC can greatly reduce the release rate.

The study in this paper addresses the preparation of directly compressed tablets with fine particle HPC matrices. The feasibility of achieving robust HPC formulations with comparable manufacturability, compactibilty, flow properties and release kinetics to formulations prepared by wet granulation is examined in the terms of the choice of directly compressible filler, drug solubility and concentration and polymer level. Lastly, a comparison to HPMC 2208 is made.
