Industrial chiral chromatography: The key to rapid process development and successful implementation

Preparative separations of stereoisomers are now routinely performed using chromatography techniques. Among the most widely used are high-pressure liquid chromatography (HPLC), supercritical fluid chromatography (SFC) and simulated moving bed (SMB). Multi-column processes can be optimized by chromatographic process simulation software, and new multi-column processes such as Varicol have been developed. This white paper by Novasep Inc. reviews state-of-the-art chromatographic processes applicable to the production of pure stereoisomers, and describes a method, based on the use of simulation software, to optimize and scale up preparative separations. This process development method is demonstrated for the separation of the enantiomers of alpha-tetralol.
